Source text - English The present invention relates to a novel 5-HT4 receptor agonist, more specifically a novel diaminopyrimidine derivative or its pharmaceutically acceptable salt having an activity as a 5-HT4 receptor agonist, a process for the preparation thereof, a pharmaceutical composition comprising the same, and a use thereof.
BACKGROUND ART
Serotonin (5-hydroxytryptamine, 5-HT), one of the neurotransmitters, is broadly distributed throughout human body including both the central nervous system and the peripheral nervous system. Approximately 95% of the human body's total serotonin is found in the gastrointestinal tract, while about 5% thereof is found in the brain. Serotonin receptors are located in intestinal nerves, enterochromaffin cells, intestinal smooth muscle, immune tissues, etc. Serotonin receptor subtypes include 5-HT1, 5-HT2, 5-HT3, 5-HT4, 5-HT5, 5-HT6, and 5-HT7. Interactions between these various receptors and serotonin are linked to various physiological functions. Therefore, various researches have been performed for developing therapeutic agents that are capable of interacting with a specific serotonin subtype as a target. The researches include identification of 5-HT4 receptors and active agents interacting therewith (Langlois and Fischmeister, J. Med. Chem. 2003, 46, 319-344).
It has been found by the previous literatures that 5-HT4 receptor agonists are useful for treating an abnormal gastrointestinal motility, i.e., dysfunction in gastrointestinal motility.
The abnormal gastrointestinal motility may result in various disorders, for example irritable bowel syndrome (IBS), constipation, dyspepsia, delayed gastric emptying, gastroesophageal reflux disease (GERD), gastroparesis, post-operative ileus, intestinal pseudo-obstruction, drug-induced delayed transit, etc.
Representative 5-HT4 receptor agonists disclosed in prior arts include tegaserod (an aminoguanidine derivative, US5,510,353), prucalopride (a benzofuran carboxamide derivative, EP0445862), cisapride (a benzamide derivative, US4,962,115), mosapride (EP0243959), etc. These compounds are known as an agent stimulating gastrointestinal motility.
DETAILED DESCRIPTION OF THE INVENTION
TECHNICAL PROBLEM
The present inventors found that a certain diaminopyrimidine derivative functions as a 5-HT4 receptor agonist, and therefore can be usefully applied for preventing or treating dysfunction in gastrointestinal motility.

Нарушения моторной функции желудочно-кишечного тракта могут привести к развитию различных болезней желудка и кишечника, например таких, как синдром раздраженного кишечника (СРК), запор (констипация), диспепсия, замедленное опорожнение желудка, гастроэзофагеальная рефлюксная болезнь (ГЭРБ), гастропарез, послеоперационная непроходимость кишечника, псевдообструкция кишечника, индуцированное лекарственными средствами замедление транзита пищи по желудочно-кишечному тракту и тому подобных.
К представителям агонистов 5-НТ4-рецепторов, раскрытых в известном уровне техники, относятся тегасерод (производное аминогуанидина, патент США US 5510353), прукалоприд (производное бензофуранкарбоксамида, патент EPO 0445862), цизаприд (производное бензамида, патент США 4962115), мозаприд (патент EPO 0243959) и тому подобные. Эти соединения известны как агенты, стимулирующие моторику желудочно-кишечного тракта.

Also an absorbing article is proposed comprising a liquid-permeable upper layer, an absorbing core and a liquid-impermeable lower layer that contains the specified non-woven fabric with the said applied ink composition. The application of ink on the non-woven fabric can be carried out by the method of flexography or a method of ink-jet printing. Printed non-woven fabric has high resistance to abrasion even in case of its contacting with fatty substance.
