I just can't find the definition of this word!

cogniscant means to have knowledge about something. In your excerpt, it means that Diderot sent a letter to Falconet in St. Petersburg in the knowledge that The Empress would also read it. By doing this, he purposely influenced the Empress to have a negative view of Falconet because he himself was no longer a friend of Falconet's.

...safe in the knowledge that...
would be the closest "layman's" terms.
